SummaryEndoplasmic reticulum, Golgi apparatus, and transit vesicles, the organelles involved in intracellular transport of storage proteins, were isolated from developing pea cotyledons. After organelle lysis and washing of the membranes with salt to remove the bulk of the matrix proteins, additional proteins were eluted from all three membrane fractions with N-acetylglucosamine. A N-acetylglucosamine-binding, 67,000 Mr integral membrane protein was released from all three membrane fractions, as well as protein body membranes, by treatment with Triton X-100 and partially purified by affinity chromatography.
