    The characteristics of a reaction involving pyrogallol and compounds related to glyceric acid in concentrated sulfuric acid were investigated. The reaction yielded a violet chromogen which exhibited an absorption maximum at 570 mμ. Among compounds reacting to form the chromogen, the following relative rates were observed: glycolaldehyde > glyceric acid > tartaric acid > 2-phosphoglyceric acid = 3-phosphoglyceric acid > 2,3-diphosphoglyceric acid.Optimal conditions for determination of monophosphoglycerates and 2,3-diphosphoglyceric acid are described. Specificity is increased by preheating the glycerates in sulfuric acid before addition of pyrogallol; contributions from the more rapidly reacting nonphosphorylated compounds are thereby reduced.Glyoxylic acid reacted with pyrogallol to produce a blue chromogen with an absorption maximum near 600 mμ.The colorimetric tests could be used for determinations of from 0.05 to 0.5 μmole of the acids under the conditions described.
