The Sulfur (VI) Fluoride Exchange reaction (SuFEx) developed by Sharpless and co-workers in 2014, is a promising new click reaction that rests on the extraordinary elevated reactivity of sulfonyl fluorides and fluorosulfates with desired appropriate nucleophiles including silyl ethers and amines. SuFEx click chemistry has various important advantages over the other previously published click reactions in that it is oxygen and water friendly, UV, and thermally inert. This peculiar reactivity of SuFEx has drawn major interest for its easy manipulation, high yields, fast reaction rates, and high tolerance towards various functional groups. This review comprehensively focuses on the sole reactivity of SuFEx click reaction and its role in the building of novel sterically hindered amides, fluorosulfates, diverse compounds from SOF4 hub, high-molecular weight polymer materials (polysulfates and polysulfonates) and surface coatings. We have also highlighted the SuFEx part in post-polymerization reactions, late-stage drug functionalization (LSF) and oil/water separations. Finally, the potential application of the SuFEx for bioconjugation is also analyzed.
