Publisher SummaryThis chapter describes procedures for labeling human interferon (Hu-IFN)-α with 125I-labeled Bolton–Hunter reagent and the purification of the radiolabeled ligand. Radioactive derivatives of IFN have been prepared by several iodination procedures, which chemically add a radioactive iodine atom or iodine-containing group. Internal radiolabeling of IFN has also been accomplished with radioactive amino acids. 125I-labeled Bolton–Hunter reagent is recommended for the iodination of Hu-IFN-α. The reaction of IFN with this reagent takes place under very mild conditions and does not reduce antiviral activity. Although the radiospecific activity of IFN with this procedure is in general less than that obtained with chloramine T or enzymobeads, it does provide a 125I-labeled IFN preparation, which is stable for up to two months. In addition to Hu-IFN-α2, Hu-IFN-αA, and Hu-IFN-α, Hu-IFN-αD can also successfully be radiolabeled with this protocol.
