Publisher SummaryThis chapter describes codeine phosphate, which occurs in two forms: one containing 1/2 molecule of water of crystallization and the other containing 1 1/2 molecule of water of crystallization. It is odorless and has a bitter taste. The chapter discusses the stereochemistry of codeine phosphate; codeine possesses five different asymmetric centers (at C5, C6, C9, C13, and C14), but because the bridged ring system imposes some rigidity upon the system, the theoretical number of optical isomers is limited to sixteen. The chapter also discusses the physical properties, biosynthesis, metabolism, and methods of analysis of codeine phosphate. The stereochemical configuration of the codeine molecule is determined by a two-dimensional study of codeine hydrobromide dihydrate. A three-dimensional study of the salt has also been carried out. The ultraviolet (UV) spectrum of codeine phosphate in water was scanned from 200 to 400 nm using Varian with 119 spectrophotometer. It exhibits a characteristic UV spectrum with a minimum at 284.8 nm.
