The assimilation of sulfur amino acid derivatives by Candida utilis and by Saccharomyces cerevisiae have been studied. The culture supplements included d-methionine, d- and l-homocysteine, dl-2-hydroxy-4-methylmercaptobutyric acid, dl-N-acetylmethionine, dl-alanyl-dl-methionine, dl-2-amino-4-(methylthio)-1-butanol (methioninol), dl-2-methylmethionine, dl-homomethionine, d- and l-cysteine, dl-allo-cystathionine, S-methyl-l-cysteine, and S-ethyl-l-cysteine. None of them caused significant inhibition of growth. Their conversion into methionine or influence on the endogenous synthesis of methionine was indicated by an increase in the level of S-adenosylmethionine. Isolation of the S-adenosylsulfonium fraction, hydrolysis, and paper Chromatographic identification of the amino acid component showed that S-adenosylmethionine was the principal product in all experiments except with S -ethyl-l-cysteine as culture supplement which led to S-adenosylethionine. S-Adenosyl-2-methylmethionine was formed in small quantity from dl-2-methylmethionine. Deamination of S-adenosylmethionine with nitrous acid gave S-inosinyl-2-hydroxy-4-methylmercaptobutyric acid. The isolation and analytical procedures applied to the biosynthetic material were tested with this compound.
