Publisher SummaryThis chapter discusses the physical properties, synthesis, stability, metabolism and pharmacokinetics, and method of the analysis of phenylpropanolamine hydrochloride. Phenylpropanolamine hydrochloride belongs to the sympathomimetic amine class of drugs and is structurally related to ephedrine hydrochloride. Phenylpropanolamine hydrochloride is widely used as a decongestant and an anorectic agent. Phenylpropanolamine hydrochloride is prepared by reacting benzaldehyde with nitroethane in 95% ethanol in the presence of sodium hydroxide to form α-(1-nitroethyl)benzyl alcohol and then reducing this nitro-alcohol to the corresponding amino compound. A stream of hydrogen chloride passed into a suitable solution of the base yields the hydrochloride. The crystal structure has been determined using single crystals of phenylpropanolamine hydrochloride obtained by slow evaporation of an aqueous solution at room temperature. Phenylpropanolamine hydrochloride has a monoclinic crystal system with possible space groups of P2l or P21/m.
