    6-Sulfanilamidoindazole (6-SAI) is an orally administered antibacterial sulfonamide which induces periarthritis in the ankles and hind paws of older rats. It is weakly or nonarthritogenic in young rats. It enhances endotoxin lethality in aged rats and produces a “synergistic toxicity” when administered in combination with some drugs. Toxicity studies in rats 10 to 12 months and 1 to 1 12 months of age demonstrated that aging was associated with enhanced colchicine lethality in control animals. Five oral administrations of 6-SAI (250 mg/kg) to groups of older rats induced inflammation in the ankles and hind paws of approximately 75% of the animals and were associated with a greater than threefold reduction in the LD50 dosage fo colchicine and greater than sixfold reduction in the LD50 dosage of vinblastine. Fibrin thrombi could be demonstrated in the renal glomerular capillaries of some aged 6-SAI pretreated (but not control) rats dying following colchicine injection. Phenylbutazone treatments suppressed the arthritis but did not inhibit colchicine lethality in 6-SAI-treated rats. Five doses of 6-SAI did not enhance colchicine lethality in young rats. Failure of 6-SAI to reduce the LD50 dosage of colchicine in young rats could not be explained by poor drug absorption.
