    A direct competitive chemiluminescent enzyme-linked immunosorbent assay (CL-ELISA) based on polyclonal antibodies with enhanced chemiluminescent detection of DIPROPYL PHTHALATE (cas 131-16-8) (DPrP) has been developed. Hapten with an amino linker introduced on the target molecule was synthesized. Bovine serum albumin was attached to the hapten by a diazotization method to prepare the immunogen. Rabbit antiserum with a sufficiently high titer to provide the determination of targeted compound was obtained. Assay conditions, including the concentration of antibody, dilution ratio of enzyme conjugate, incubation time, the effects of buffer concentration and pH, and so on, were optimized. Under the optimized conditions, the 50% inhibition values of 0.19 μg/L for DPrP was achieved with a limit of detection of 0.03 μg/L. Little cross-reactivity (<9%) to structurally related phthalates was observed. The developed method has been used to quantify DPrP in water and milk samples without purification or preconcentration steps. The recovery of DPrP ranged from 86.4% to 119.5% at three concentrations (1, 10, and 50 μg/L), and the coefficients of variation were less than 14%. The results suggested that the developed CL-ELISA would be very suitable for rapid monitoring of DPrP in food samples.
