Publisher SummaryThis chapter provides an overview of captopril. It discusses the name, formula, molecular weight, and appearance, color, and odor of captopril. Various physical properties are discussed— spectral properties, solid state properties, and solution data. Several analytical tests and methods—elemental analysis, spectrophotometric methods, chromatographic methods, and titrimetric methods are reviewed. Captopril in aqueous solution undergoes an oxygen facilitated, first order, and free radical oxidation at its thiol to yield captopril disulfide. Transition metal ions most effectively catalyze oxidation of captopril through a recycling of oxygen free radicals. The greater the captopril concentration, the slower is the oxidation. Various chromatographic methods are discussed in the chapter—thin layer radiochromatography (TLRC), gas chromatography-mass spectroscopy (GC-MS), gas chromatography-flame photometric detection (GF-FPD), spectrofluorometry, radioimmunoassay (RIA), and semiautomated Ellman colorimetry. Development and application of a simple, highly sensitive RIA for N-ethylmaleimide (NEM)-captopril in blood plasma is described. The lower detection limit is two ng per ml of plasma.
