The invention relates to a synthetic method of (3aS, 6aR)-1, 3-dibenzyl tetrahydro-4H-thieno[3, 4-d]imidazole-2, 4-(1H)-dione. According to the synthetic method provided by the invention, the raw materials are cheap and easily available, and the method is simple in reaction and easy to operate. Mannich reaction is catalyzed by a chiral catalyst to construct a beta-position chiral center and an alpha chiral center is constructed by SN2 substitution reaction and thermal rearrangement of the alpha-position, so that the synthetic method is low in cost and suitable for industrial production.
