1. The brief introduction of Saikosaponin a (SSa)

Saikosaponin a (SSa) is β-D-Galactopyranoside, (3β,4α,16β)-13,28-epoxy-16,23-dihydroxyolean-11-en-3-yl 6-deoxy-3-O-β-D-glucopyranosyl-. Saikosaponin a (SSa), as a principal active ingredient in the root of Bupleuri Radix, exhibits antiinflammatory, hepatoprotective, antitumoral, and antiviral activities. Saikosaponin a (SSa) and its epimer saikosaponin d (SSd) are major triterpenoid saponin derivatives from Radix bupleuri (RB), which has been long used in Chinese traditional medicine for treatment of various inflammation-related diseases.

The study investigated the effects of EGb761 and SSa on carcinogenesis and apoptosis in HCC cells. In the present study, the anti-inflammatory activity, as well as the underlying mechanism, of SSa and SSd was investigated in lipopolysaccharide (LPS)-induced RAW264.7 cells.

2. Saikosaponin A and D

Saikosaponin A [20736-09-8] and D [20874-52-6] in 0.2% KOH-MeOH exts were quant. Several substances with their cas registry numbers 20874-52-6 and 58558-08-0 may be changed into the corresponding B1 [58558-08-0] and B2 [58316-41-9] by mild acid. Saikosaponin B1 and B2 were detd by HPLC with a reversed-phase column (C18-10) and a mobile phase MeOH-H2O-AcOH-Et3N (:0.2:0.2). This method was applied to produce Daisaikoto and Shosaikoto. Saikosaponin A and D in Daisaikoto were 136-472 mg/g and 7/-320 mg/g, and those in Shosaikoto were 171-385 mg/g and 113-430 mg/g, resp.

3. Effect of saikosaponin-A

Effect of saikosaponin-A is a triterpenoid glycoside, isolated from Bupleurum falcatum on experimental allergic asthma. Someone had done a reasearch and got some results indicated that SSA had an inhibitory activity against allergic asthma. This activity seems to derive from both antagonism of the histamine action and inhibition of allergic mediators. Additional mechanisms may also be involved.
