Taps, n-tris (hydroxymethyl) methyl-3-aminopropanesulfonic acid, is an amphoteric buffer, which is widely used in biochemistry and molecular biology. It is a part of Tris series buffer, pH buffer range 7.7-9.1, soluble in water (25g / 50ml). Taps, as a buffer system commonly used in DNA screening system, can also be used as a buffer component of RNA samples. It is suitable for electron transfer and phosphorylation research of chloroplast thin layer preparation. It can protect oxyhemoglobin from oxidation to methemoglobin during freeze-drying. It can also be used as a background electrolyte for protein microanalysis by capillary zone electrophoresis.
At present, there are few reports on the synthesis of taps. Chen gui'e and others have studied the synthesis of taps in detail. Tris and 1,3-propanesulfonic acid lactone (1,3-ps) were used as raw materials to react in alcohol solvent
1. Weigh equal moles of Tris and 1,3-ps, and take appropriate amount of alcohol solvent into a 250ml three port flask with condensation reflux. Under the condition of stirring, heat up and reflux;
2. Stop the reaction after a period of time; At room temperature, after the reaction solution is cooled to room temperature, the tap filter cake is washed 2 ~ 3 times with an appropriate amount of anhydrous ethanol, then the tap filter cake is taken out and dried in a vacuum drying oven, and the filtrate is recovered for use.

In the above synthesis process, the reaction time, different alcohol solvents and the recycling of reaction solvents have some effects on the yield and purity of taps. The results showed that the highest yield was 70.3% when the reaction time was controlled at 6 hours; For ethanol, n-propanol, isopropanol and n-butanol, the yield and purity of taps with n-butanol as reaction solvent were 70.3% and 94.36%, respectively; In the reaction process, the recycling of reaction solvent is conducive to improve the yield and purity of taps.
In conclusion, taps was synthesized from Tris and 1,3-ps, the reaction time was controlled at 6 hours, n-butanol was used as solvent, and recycled in the reaction process, the yield and purity of taps were the best.
