Determination of heparin

Heparin is a substance with anticoagulant activity. The higher the content of heparin sodium in solution, the stronger the anticoagulant effect. The potency of heparin sodium produced in China is mostly determined by sheep plasma method. The principle of this method is to add standard and test sample into sheep plasma, and observe the coagulability of standard and test sample heparin sodium added into human tube after a certain time of heavy calcification of sheep plasma. If the standard and test sample are mixed into the same concentration and get the same coagulability, then their potency is the same, so as to compare and determine the potency of unknown sample. The following is the procedure of detecting heparin titer in sheep plasma.

Heparin sodium powder

1. According to the estimated potency, weigh the heparin powder to be tested;

2. Add 0.9% normal saline to dissolve heparin sodium powder, and fix the volume to 100 ml;

3. Take out 2 ml of heparin solution after constant volume, and fix the volume to 50 ml to get the solution to be tested;

4. Take out two groups of test tubes, 10 tubes in each group, and inject 110 μ l to 200 μ l of standard heparin solution and solution to be tested into two groups respectively (to avoid mixing, mark the amount added on the test tube);

5. 20 μ l 8% sodium citrate solution was added to each tube, and 0.8 ml 25% calcium chloride was added;

6. After thawing and filtering, 1 ml of sheep plasma was added into each tube;

7. Cover each tube with a rubber plug, turn it over for 3-4 times, mix well, and make the inner wall wet;

8. The heparin solution volume corresponding to 1 / 2 freezing point of the standard sample and the sample to be tested was recorded and read.

The formula for calculating the potency of the sample to be tested is as follows:

F = estimated potency of sample to be tested x microliter of 1 / 2 freezing point of standard ÷ Microliter of sample 1 / 2 freezing point
