How is heparin extracted

Heparin is a kind of mucopolysaccharide sulfate which is composed of glucosamine, l-iduloside, n-acetylglucoside and D-glucuronic acid. Its average molecular weight is 12000-15000. Heparin has strong negative charge, which can strengthen antithrombin - Ⅲ (at - Ⅲ) to inactivate serine protease, thus it has many functions, such as preventing thrombin formation, antithrombin and platelet aggregation. Therefore, according to the above principles, heparin sodium is commonly used as blood anticoagulant in blood test.

How is heparin sodium produced

At present, heparin sodium is mainly produced by enzymatic hydrolysis, high temperature inactivation and resin purification of crude heparin. Due to insufficient enzymatic hydrolysis, resin adsorption is incomplete and the product yield is low. In the process of salt elution, the amount of salt is very large, the cycle is long, and more wastewater is produced, which pollutes the environment. At the same time, due to insufficient enzymolysis and more impurities, the regeneration times of the resin are increased, the use cost of the resin accessories is increased, the service life of the resin is reduced, and the production cost is increased.

The other process is to hydrolyze the crude heparin, kill the enzyme at high temperature, and then oxidize it for 3-5 times, then precipitate it with alcohol repeatedly to remove the related impurities. The process is long and time-consuming, and the multiple oxidation has an impact on the structure of heparin molecule, and the yield is low.

Because of the poor stability of the enzyme and insufficient enzymatic hydrolysis, the complete extraction of heparin can not be achieved, resulting in very complex follow-up process, high production cost, long production cycle and difficult control of product quality.

At present, trypsin immobilized on magnetic materials is widely used to hydrolyze crude heparin, so that the enzyme has higher stability and better activity, and can completely release heparin from crude heparin. At the same time, it does not need high temperature to kill the enzyme and will not be converted into impurities, which makes the purification process more difficult. At the same time, trypsin immobilized by magnetic materials can be easily separated from the hydrolysate and recovered. In this way, the impurities such as protein, chondroitin sulfate and dermatan sulfate can be simply and completely removed through the follow-up process. At the same time, the recovered immobilized trypsin particles can be recycled repeatedly, which can reduce the cost of enzyme and the subsequent impurity removal process, simplify the heparin refining process, improve the production efficiency and greatly reduce the production cost
