Tris buffer, or Tris buffer, is usually used with hydrochloric acid to adjust the pH value to prepare buffer. Some of them are adjusted with acetic acid, boric acid, glycine and other acids. The pH value of Tris aqueous solution is about 10.5. Generally, hydrochloric acid is added to adjust the pH value to the required value to obtain the buffer solution with this pH value. Characteristics of Tris buffer
Trihydroxymethyl aminomethane itself is a weak base, and the aqueous solution is alkaline, so hydrochloric acid is used to adjust the buffer solution instead of sodium hydroxide. Tris molecular structure contains an amino group, which can react with aldehydes. Therefore, the use of Tris buffer in the system containing aldehydes may be affected.
Tris buffer is suitable for nucleic acid related experiments
Tris buffer is usually neutral and weakly alkaline. DNA nucleic acid molecules will be deprotonated in such a solution to improve the solubility. Therefore, Tris can be used as a good solvent for nucleic acid (and also suitable for protein). In nucleic acid related experiments, Tris is preferred as a buffer.
