Determination of heparin sodium titer in sheep plasma

Heparin is a substance with anticoagulant activity. The greater the content of heparin sodium in the solution, the stronger the anticoagulant effect. The titer of heparin sodium products produced in China is mostly determined by sheep plasma method. Its principle is to add the standard and test sample into the sheep plasma, and observe the coagulation degree of the standard and test sample heparin sodium added into the human tube after waiting for a certain time for the re calcification of the sheep plasma. If the standard and test sample are mixed into the same concentration and get the same coagulation degree, it indicates that their titers are also the same, so as to compare and determine the titers of unknown samples. The following are the steps of detecting heparin titer in sheep plasma.

Heparin sodium powder

1. Weigh the heparin powder to be tested according to the estimated titer;

2. Add 0.9% normal saline to dissolve heparin sodium powder and fix the volume to 100ml;

3. Take out 2ml of constant volume heparin solution, and fix the volume to 50ml to obtain the solution to be tested;

4. Take out two groups of test tubes with 10 tubes in each group. The standard heparin solution and the solution to be tested injected into the two groups are 110 μ l to 200 μ L respectively (to avoid mixing, the amount added can be marked on the test tube);

5. Add 20 μ l of 8% sodium citrate solution and 0.8 ml of 25% calcium chloride to each tube;

6. Add 1ml of thawed and filtered sheep plasma into each tube;

7. Wet the inner wall of each test tube for 3-4 times;

8. Put it vertically into the water at 38 ℃ for 1 hour, take it out, observe the solidification degree of each tube, record the volume of heparin solution corresponding to 1 / 2 freezing point of standard sample and sample to be tested, and read it.

F = estimated potency of sample to be tested x 1 / 2 freezing point microliter of standard product ÷ 1 / 2 freezing point microliter of sample
