HEPES is a buffer reagent for cell culture. Its buffer range is 6.8-8.2. In open cell culture conditions, its pH value is often affected by the outside world. If HEPES buffer is added, it can prevent the increase of pH oxidation in the culture medium. Therefore, HEPES buffer is often indispensable in the process of cell culture. In addition, it is also widely used in the uptake of organic molecules, the change of oxidative stress mechanism or the inhibition of ions.

Compared with bicarbonate, hepe buffer has higher solubility and more stability, and has the smallest complexation with metal ions. Under different use conditions, it is proved that HEPES can be responsible for the significant modification of biotechnology, the absorption and other parameters of organic molecules, and there are several molecular mechanisms for the inhibition of oxidative stress mechanism and ion channel.
Prions, also known as prions, protein infectious factors, prions or infectious proteins, are a kind of small molecule non immune hydrophobic proteins that can infect animals and replicate in host cells. In the process of seeking prion gene therapy in our laboratory, we infected lentiviruses on cells with prion in the supernatant of HEK 293T containing related proteins. In the control experiment, a significant inhibition of the accumulation of prion protein (PrPSc) was observed by using unrelated lentivirus or culture medium alone. The abnormal subtype of PrPSc was finally confirmed to be related to the presence of hepe in the culture medium.
When HEPES supplemented cell culture medium was used on prion infected cells, there was a significant concentration dependent inhibition of the accumulation of prion protein abnormal subtype (PrPSc). This effect exists only in living cells and has not been confirmed. It is considered to be related to changes in PRP environment or biology. This inhibition of HEPES can also be used to prevent prion contamination or reproduction in cell culture.
