Atovaquone is hydroxy 1,4-naphthoquinoline, which is a homologue of Coenzyme Q and has the activity against several protozoa. For Plasmodium, the site of action is the cytochrome bcl binding site (binding site III). Atovaquone can reversibly bind to the 11500Da molecular group on the polypeptide. Dihydroorotate dehydrogenase is an important enzyme in the biosynthesis of pyridine. Coenzyme Q connects mitochondria for electron transfer. Therefore, this product prevents the synthesis of pyridine by inhibiting electron transfer. Some metabolic enzymes participate in the electron transport of mitochondria through Coenzyme Q. Therefore, the inhibitory effect of this product on electron transport actually inhibits the activity of these enzymes.
Atovaquone is apply for oral treatment of AIDS patients suffering from mild to moderate KF pneumonia who cannot tolerate SMZ-TMP.
