Tris, fully known as trimethylaminomethane, belongs to one of biological buffers. The appearance is white crystalline powder with good water solubility. It is soluble in ethanol, acetic acid and ethyl ester, but insoluble in ether and carbon tetrachloride. Tris can be configured as a solution. Tris buffer is a kind of solution that can resist the influence of a small amount of strong acids and bases outside and maintain the pH value of the system unchanged. Tris buffer range:
Tris is a weak base, and its pKa value is 8.1 at room temperature of 25 ℃. According to the buffer theory, the effective buffer range of Tris buffer is between pH 7.0 and 9.2, and the pH of 0.1mol/l aqueous solution is 10.4. Generally, hydrochloric acid is added to adjust the pH value to obtain the buffer with the required pH value. But at the same time, attention should be paid to the effect of temperature on its PKA and Tris buffer. For every 1 ℃ decrease in temperature, the pH increases by about 0.03 units, and for every 10 times dilution of concentration, the pH decreases by 0.03-0.05 units. Moreover, Tris buffer is very easy to absorb CO2 in the air, so the prepared buffer needs to be sealed. In addition, Tris buffer will interfere with some pH electrodes, so electrodes compatible with tris buffer need to be used.
Application of Tris in buffer:
1. 1m tris-hcl6.8, 1.5m tris-hcl8.8 and 5xtris glycine electrophoresis buffers are the most commonly used reagents for SDS-PAGE.
2. EDTA was added to Tris hydrochloric acid buffer to make te buffer. Te buffer was used for DNA stabilization and storage. Tae buffer can be obtained by replacing hydrochloric acid with acetic acid, and tbe buffer can be obtained by replacing boric acid. Both TAE and tbe buffer are commonly used in nucleic acid electrophoresis experiments.
3. Tris was used for protein crystal growth under different pH conditions.
4. The low ionic strength of Tris buffer can be used for the formation of intermediate fibers of nematode lamin.
