Synthesis route of Luminol
The chemical name of luminol is 3-aminophthaloyl hydrazide, CAS No.: 521-31-3, which can be prepared from 3-aminophthalic acid. The raw material is dehydrated to produce 3-Nitrophthalic anhydride, which can be used for organic synthesis and alcohol determination. After nitration, both 3-nitro-phthalic acid and 4-nitro-phthalic acid can be obtained. After the reaction of the former product with hydrazine (hydrazine), 3-aminophthaloyl hydrazide, namely luminol, can be prepared; The other can produce isoluminol.

Separation of luminol intermediate by-products
According to the synthesis route of luminol, there is a by-product in the intermediate, which will react with hydrazine to produce isoluminol, so it is necessary to remove the by-product. In 3-nitro-phthalic acid molecule, nitro has a great impact on the ortho carboxyl group. It will form an intramolecular hydrogen bond with carboxylic acid. In addition, the intramolecular hydrogen bond between adjacent dicarboxylic groups will significantly inhibit the dissociation of the whole carboxylic acid molecule, resulting in the decrease of its water solubility. In 4-nitro-phthalic acid, it is difficult to form intramolecular hydrogen bond between nitro and carboxylic acid. Therefore, its dissociation degree in water is relatively larger and its water solubility is better. The isomers produced after nitration of phthalic anhydride have different water solubility, which can be used to remove by-products.
Use of luminol luminescent reagent
Luminol belongs to enzymatic chemiluminescence. The detection needs to be catalyzed by peroxidase HRP, and then hydrogen peroxide can be added under alkaline conditions to stimulate the chemiluminescence reaction; HRP can label proteins or antibodies, so it can be combined with immune response.
