Trimethylolaminomethane buffer, i.e. Tris buffer, is usually mixed with hydrochloric acid to adjust the pH value to prepare buffer, and some are adjusted with acetic acid, boric acid, glycine and other acids, while many other kinds of buffers such as bicine, mops, HEPES and caps are adjusted with sodium hydroxide alkali solution, which is the unique feature of Tris buffer.

Basic properties of trimethylolaminomethane
At room temperature (25 ℃), the pKa of Tris is 8.1; according to the buffer theory, the effective buffer range of Tris buffer is between pH 7.0 and 9.2. The pH of Tris aqueous solution is about 10.5. Generally, hydrochloric acid is added to adjust the pH value to the required value to obtain the buffer of this pH value. The pKa of Tris is greatly affected by temperature, which will lead to the change of pH value. Attention should be paid to the influence of temperature.
Characteristics of Tris buffer
Trimethylolaminomethane itself is a weak base, and the aqueous solution is alkaline. Therefore, hydrochloric acid is used to adjust the buffer rather than sodium hydroxide. Tris molecular structure contains an amino group, which can condense with aldehyde. Therefore, the use of Tris buffer may be affected in the system containing aldehyde.
Tris buffer is suitable for nucleic acid related experiments
Tris buffer is usually neutral and weak alkaline. DNA nucleic acid molecules will be deprotonated in such a solution to improve the solubility. Therefore, Tris can be used as a good solvent for nucleic acid (also suitable for protein) , in nucleic acid related experiments, Tris is preferred as buffer.
