Dithiothreitol is a commonly used reducing agent, also known as dimercaptothreitol (DTT for short), which has strong reducibility, and its reducibility is largely due to the conformational stability of its oxidation state six-membered ring (containing disulfide bonds). The reducing power is affected by the pH value, and can only play a reducing role when the pH value is greater than 7. This is because only the deprotonated thiol anion (-s -) is reactive, while the thiol (-SH) is not. The pKa of the sulfhydryl group is usually ~ 8.3.
