An intravenous injection of the contrast medium Meglumine Ioglycamide (Biligram) led to a fatal incident in a patient (E.O.) suffering from Macroglobulinemia Waldenström. Subsequently the interaction of IgME.O. with mono- and divalent drugs bearing the antigenic determinant 3-amino-2,4,6-triiodobenzoic acid was investigated, giving evidence for an antigen-antibody-like reaction between IgME.O. and loglycamide as well as related compounds. The precipitin reaction of IgME.O. and the divalent drug Iodipamide was studied in detail. In the region of antibody excess, linear increasing amounts of divalent hapten led to an exponential increase of the association constant. A positive cooperation between the antigen binding sites within the IgM molecule in the course of multiple binding of identical antigenic determinants is discussed.
