PRIMYCIN (cas 113441-12-6), an antibiotic isolated from a species ot Actinomycetes, inhibits the growth of Euglena gracilis and Astasia longa, but adaptation occurs after a lag whose duration depends sensitively on primycin concentration. A low concentration of actinomycin D, which by itself scarcely inhibits growth of these cells, enhances the growth inhibition caused by primycin. Low concentrations of primycin prevent formation of an induced acid phosphatase. The inhibitory effect of primycin can be prevented by heat-killed cells and by DNA, RNA, and polynucleotides. Primycin interacts with polynucleotides in vitro by forming easily sedimentable aggregates under certain conditions. Primycin does not appreciably affect the melting curve of DNA, but it prevents the return of hypochromism on cooling the heated DNA. These results suggest that this antibiotic inhibits growth by interacting with the nucleic acids of the cells, perhaps preferentially with the single stranded forms.
