The effect of polyvinylpyridine N-Oxide and polyvinylpyrrolidone on quartz cytotoxicity was studied in guinea pig macrophages using leakage of acid phosphatase enzyme through the lysosomal membrane as a parameter of the cytotoxicity of quartz dust. The macrophages were either pretreated with the polymers before being exposed to quartz or the animals from which the macrophages were harvested were pretreated with them. The studies were extended on macrophages harvested from normal animals and inoculated in vitro with uncoated quartz, quartz coated with polyvinylpyridine N-oxide and quartz coated with polyvinylpyrrolidone.Pretreatment of cultures with polyvinylpyridine N-oxide before quartz exposure and coating of quartz with the polymer prevented almost completely the leakage of acid phosphatase enzyme. Pretreatment of animals with polyvinylpyridine N-oxide delayed and inhibited to a large extent the occurrence of these changes. Polyvinylpyrrolidone prevented leakage of acid phosphatase enzyme only when used for coating the quartz surface but not when the animals or the cells were pretreated with it.
