SummaryA 2-phosphoglyceric acid phosphatase (2-phosphoglycerate phosphohydrolase) has been identified and partially purified from beef liver. The purification of 2-phosphoglyceric acid phosphatase from other non-specific liver phosphatases was based on the relative activity of 2- or 3-phosphoglycerate to α-glycerophosphate and β-glycerophosphate during the preparation. The final preparations had high specificity for 2-phosphoglycerate but with variable low activity with other substrates. Further purification was prevented by low final yields of enzyme. Data are presented on the kinetics, specificity and inhibition of the enzyme preparations obtained. The identification of 2-phosphoglyceric acid phosphatase completes the description of the mammalian enzymes of serine biosynthesis from carbohydrate intermediates. Both “phosphorylated” and “non-phosphorylated” pathways appear to be present in mammalian systems but the relative significance of each is not yet known.
