Tris (hydroxymethyl) methyl-3-aminopropanesulfonic acid English name taps, CAS No. 29915-38-6; Molecular weight 243.28; Molecular formula: c7h17no6s; This product is a white crystalline powder, soluble in organic solvents such as water, and the pH buffer range is 7.7-9.1.
Synthesis process of taps:
The synthesis process of taps uses Tris and 1,3-propanesulfonic acid lactone (1,3-ps) as raw materials to react in alcohol solvent. The operation process is as follows:
1. Weigh the same molar amount of Tris and 1,3-ps, and take an appropriate amount of alcohol solvent into a 250ml three mouth flask with condensation reflux. Under stirring, heat up and reflux;
2. Stop the reaction after a period of time; At room temperature, after the reaction solution is cooled to room temperature, it is filtered under reduced pressure. The taps coarse filter cake is washed with an appropriate amount of absolute ethanol for 2 ~ 3 times. The taps filter cake is taken out and dried in a vacuum drying oven. The filtrate filtered under reduced pressure is recovered for use.
Solution preparation method:
When taps is prepared into buffer solution, sodium hydroxide alkaline solution is usually used to adjust the pH value.
Firstly, weigh the mass of taps powder and sodium hydroxide solid (the relative molecular mass of taps is 243.28 and that of sodium hydroxide is 40), then prepare the solution respectively and mark liquid a and liquid B.
If the reaction system needs to eliminate sodium ions, potassium hydroxide is used instead of sodium hydroxide, and the temperature is controlled at room temperature 20 ℃ when preparing the solution. If you need to prepare a buffer system with more accurate pH value, you can prepare it according to the above method, then measure it with a pH meter, and then slowly add liquid a or liquid B for adjustment.
usage method:
Completely dissolve the weighed taps reagent powder with deionized water. Pay attention not to add too much water at the beginning of dissolution to ensure all dissolution. Finally, continue to add water to fix the volume. Attention shall be paid to the temperature during constant volume. If the temperature changes too much due to exothermic or endothermic heat during dissolution, the constant volume shall be determined when the temperature is close to room temperature. After configuration, seal the cap of the volumetric flask and keep it for standby at any time. Mark taps solution a, concentration, configuration date and other information.
Application of Taps:
1. Taps, as a common buffer system in DNA screening system, can also be used as a buffer component of RNA samples. It is suitable for the study of electron transfer and phosphorylation of chloroplast thin layer samples
2. Protect oxygenated hemoglobin from oxidation to methemoglobin during freeze-drying
3. It can be used as background electrolyte for protein microanalysis by capillary zone electrophoresis.
4. It can be used as an important pH stabilizer, which is generally mixed with weak acid and its conjugated base to obtain appropriate pH value.
5. It is usually used in some biochemical diagnostic kits, PCR diagnostic kits and DNA / RNA extraction kits.
